The actor - best known for playing Desmond Hume on Lost - will appear in the episode 'Letters of Transit'.
"It's similar to Lost in that it's high-stakes life-and-death stuff… a very exciting episode to work on," Cusick told TVLine.
Australian actress Georgina Haig has also been cast in the instalment, which is set in the year 2036 in a break from the usual Fringe format.
"I do most of my scenes with Georgina and John Noble (Walter Bishop), who is a phenomenal actor, and it was a great experience," Cusick explained.
Joshua Jackson (Peter Bishop) previously implied that 'Letters of Transit' could set up a fifth season of Fringe, with Cusick hinting that he could return to the show in future episodes.
"That show is like Lost in that it can go any which way, so yes, [I'd come back]," he said.
